[Intended Use]

This kit is suitable for in vitro qualitative detection of Coxsackie virus A6, A10 nucleic acids in throat swabs and herpes fluid samples of patients with Hand-foot-mouth disease (HFMD), and provides an auxiliary means for the diagnosis of these patients.
HFMD is an infectious disease caused by Enterovirus, and studies have found that Coxsackie virus A6 and A10 are associated with sporadic and explosive events of HFMD. The disease mostly occurs in children under 5 years old, and can cause herpes on the hands, feet, mouth and other parts. A few children may cause complications such as myocarditis, pulmonary edema, aseptic meningoencephalitis, etc.

[Test Principles]

This detection kit uses the PCR method combined with the in vitro amplification detection technology of fluorescent probes. Specific primers and probes are designed for Coxsackie virus A6 (CA6) and A10 (CA10) respectively, and the specific probes 5’ are marked as FAM (CA6), VIC (CA10) and ROX (internal control) fluorophores. The extracted RNA is reverse transcribed into cDNA, and during the PCR amplification, specific primers and probes bind to their respective target sequences separately. When the Taq enzyme encounters the probe bound to the target sequence, it exerts the function of 5'-end exonuclease to separate the reporter fluorophore and the quencher group, so that the fluorescence monitoring system can receive the fluorescence signal, that is, each time a DNA strand is amplified, a fluorescent molecule is formed. Accumulation of fluorescent signal is achieved in complete synchronization with PCR product formation.

The qualitative detection of Coxsackie virus A6 and A10 nucleic acids in swabs and herpes fluid samples of HFMD patients can be realized by detecting different fluorescent signal changes, which provides an auxiliary means for the diagnosis and treatment of patients with Coxsackie virus A6 and A10 infection.

[Storage Conditions and Shelf-life]

1. Unopened kits: Should be stored at a temperature not higher than 30°C and protected from light, and the shelf life is 12 months.
2. The freeze-dried nucleic acid reaction buffer and the positive control should be tested immediately after being reconstituted. The remaining reconstituted solution should be stored at -25°C to -15°C away from light, and the shelf life is 1 week and avoid the repeated freezing and thawing (should be no more than two cycles).

3. Transport condition: Can be transported stably for 1 month below 37°C.
