[Intended Use]
This kit is used for in vitro qualitative detection of Plasmodium nucleic acid in peripheral blood samples of patients with suspected Plasmodium infection.

Malaria (Mal for short) is caused by Plasmodium, which is a single-celled eukaryotic organism, including Plasmodium falciparum Welch, Plasmodium vivax Grassi & Feletti, Plasmodium malariae Laveran, and Plasmodium ovale Stephens. It is a mosquito-borne and blood-borne parasitic disease that seriously endangers human health. Of the parasites that cause malaria in humans, Plasmodium falciparum Welch is the deadliest. The incubation period of different malaria parasites is different, the shortest is 12-30 days, and the longer one can reach about 1 year. After the paroxysm of malaria, symptoms such as chills and fever may appear. The patients may have anemia and splenomegaly. The serious patients may have coma, severe anemia, acute renal failure which may lead to the death of patients. Malaria is distributed worldwide, mainly in tropical and subtropical regions such as Africa, Central America, and South America[2].

At present, there are three detection methods: Plasmodium blood smear, antigen detection, and nucleic acid detection.The detection of Plasmodium nucleic acid by isothermal amplification technology has the advantages of rapid detection and simple response, and is suitable for detection in large-scale malaria epidemic areas.

The test result of this kit is not the only indicator for the diagnosis of Plasmodium infection in patients, and must be comprehensively analyzed in combination with the clinical manifestations of patients and other test results.

[Test Principles]

This kit uses a combination of PCR amplification and fluorescent probes. Design specific primers and probes for the highly conserved region of Plasmodium, and the probes are labeled with FAM fluorescent groups. At the same time, an internal control is introduced, and a specific primer probe for fluorescence detection is designed. The 5' end of the probe is labeled with VIC fluorophore, and the 3' end quencher is BHQ2. 

In the process of PCR amplification, specific primer probes bind to their respective target sequences. When Taq enzyme encounters the probes bound to the target sequence, it exerts the function of 5′-end exonuclease, so that the reporter and the quencher is separated, and the fluorescence monitoring system can receive the fluorescent signal, that is, each time a DNA chain is amplified, a fluorescent molecule is formed, realizing the complete synchronization of the accumulation of the fluorescent signal and the formation of the PCR product. By detecting different fluorescence signal changes, realize the qualitative detection of Plasmodium nucleic acid in samples, and provide auxiliary means for the diagnosis and treatment of patients with enterovirus infection.

[Storage Conditions and Shelf-life]
The kit should be stored below -18°C protected from light. The shelf life is 12 months. The number of repeated freezing and thawing should not be more than 4 cycles.

After opening, the kit should be stored below -18°C. If it is stored 2-8°C, please use it within 1 week.

[Requirements for Samples]

1.Sample collection: whole blood, dried blood spots
Dried blood spots: Use a disposable blood collection needle to collect blood from the earlobe or the end of the finger. The baby can collect blood from the thumb or heel. And drop the whole blood on the filter paper, and seal the bag after drying. Save the whole blood sample in the micro anticoagulation tube for inspection.
Whole blood: collected EDTA anticoagulated blood.
2. Precautions for sample collection: Avoid contamination during sample collection, storage and transfer.
Sample safety: All samples are considered to be potentially infectious, and operations are performed in accordance with relevant national standards.
3. Storage
The samples to be tested should be stored at 2~8°C for no more than 7 days; stored at temperatures below -18°C for no more than 3 months; can store at temperatures below -70°C for long time. Repeated freezing and thawing should be avoided.
