[Intended Use]
This kit is used for in vitro qualitative detection of helicobacter pylori nucleic acid in gastric mucosal biopsy tissue samples or saliva samples of patients suspected of being infected with helicobacter pylori, and provides an auxiliary means for the diagnosis of patients with helicobacter pylori disease. The test results are for clinical reference only, and the final diagnosis should be comprehensively considered in close combination with other clinical indicators.

Helicobacter pylori (Hp) is a Gram-negative helical microaerophilic bacterium. Since the Australian scientists Warren and Marshall isolated Hp on the 26th April, 1982, its relationship with human health has been highly concerned by many domestic and foreign medical scholars for a long time. Hp has a global infection and is closely related to many upper gastrointestinal diseases. It is an important pathogenic factor for chronic gastritis, gastric ulcer, duodenal ulcer, and upper gastrointestinal tumors, and the World Health Organization has classified it as a class I carcinogen. With the in-depth research, it is found that Hp infection is not only related to gastrointestinal diseases, but also can cause cardiovascular and cerebrovascular diseases, hepatobiliary diseases, chronic bronchitis, iron deficiency anemia and other system diseases, and even induce tumors. Therefore, the prevention, diagnosis and treatment of Hp infection have increasingly attracted the attention of many countries and regions.

[Test Principles]

This kit uses fluorescent probe real-time PCR technology to design specific primer probes in the specific conserved region of helicobacter pylori. The reporter of the probe is FAM, the quencher is BHQ1, and the human gene is selected as internal control at the same time. Design a specific primer probe, the reporter is VIC, and the quencher is BHQ1. During the amplification process, the 5′-3′ exonuclease activity of Taq enzyme realizes the complete synchronization of the formation of PCR products and the accumulation of fluorescent signals, and realizes qualitative detection of helicobacter pylori nucleic acid in human gastric mucosa biopsy tissue samples or saliva samples.

[Storage Conditions and Shelf-life]

The kit should be stored below -18°C protected from light. The shelf life is 12 months(please use within the shelf life). After opening, the number of repeated freezing and thawing should not be more than 4 cycles.It can be stably stored for 5 days below -18°C and protected from light during transportation.

[Requirements for Samples]

1.Sample Collection: Human gastric mucosa tissue samples or saliva samples.
1.1 Human gastric mucosa tissue samples
Take paraffin-embedded human gastric mucosa tissue samples, cut 5-6 slices of paraffin tissue about 5 μm from each sample, and put them into sterile centrifuge tubes.
1.2 Saliva samples
Saliva samples are collected on an empty stomach in the early morning of the examination. Please rinse your mouth with drinking water 30 minutes before collecting a saliva sample. Press the tip of the tongue against the root of the upper or lower jaw to enrich the saliva, and gently spit the saliva into the collection funnel until the liquid saliva (non-bubble) reaches the height of the 2mL scale. Turn the collection tube upside down to thoroughly mix the saliva with the saliva preservation solution.
2. Precautions for sample collection: Avoid contamination during sample collection, storage and transfer.

Sample safety: All samples are considered to be potentially infectious, and operations are performed in accordance with relevant national standards.
3. Storage
Paraffin-embedded pathological tissue or section samples are generally stored for no more than 3 years. Saliva samples should be stored at 2-8°C for no more than 7 days, and should be stored below -18°C for no more than 2 months, and could be stored below -70°C for long-term.
