This kit is used for the qualitative detection of adenovirus nucleic acid in stool samples in vitro. Adenovirus ( Adv) belongs to the Adenovirus family. Adv can proliferate and cause disease in cells of the respiratory tract, gastrointestinal tract, urethra, and conjunctiva. It is mainly infected through the gastrointestinal tract, respiratory tract or close contact, especially in swimming pools with insufficient disinfection, which can increase the chance of transmission and cause outbreaks [1-2]. Adv mainly infects children. The gastrointestinal tract infections in children are mainly type 40 and 41 in group F. Most of them have no clinical symptoms, and some cause diarrhea in children. Its mechanism of action is to invade the small intestinal mucosa of children, making the intestinal mucosal epithelial cells smaller and shorter, and the cells degenerate and dissolve, resulting in intestinal absorption dysfunction and diarrhea. Abdominal pain and bloating may also occur, and in severe cases, the respiratory system, central nervous system, and extraintestinal organs such as liver, kidney, and pancreas may be involved and the disease may be aggravated [3-5].
The test results of this kit are not the only indicator for the diagnosis of adenovirus type 41 infection in patients. It must be combined with the clinical characteristics of the patient and other laboratory test data to correctly determine the infection of the pathogen, and formulate a reasonable treatment plan to make the treatment safe and effective.
This kit adopts fluorescent probe real-time PCR technology, selects the conserved region of adenovirus type 41 to design specific primer probes. The 5' of the probe is labeled with FAM, and the 3' quencher is BHQ1. In the process of PCR amplification, specific primers and probes bind to their respective target sequences. When Taq enzyme encounters the probe bound to the target sequence, it exerts the function of 5'-end exonuclease, making the reporter and the quencher separated, so that the fluorescence monitoring system can receive the fluorescent signal, that is, a fluorescent molecule is formed every time a DNA chain is amplified, realizing the complete synchronization of the accumulation of the fluorescent signal and the formation of the PCR product. At the same time, endogenous internal control is introduced to avoid false negative in the test.
[Storage Conditions and Shelf-life]
The kit should be stored below -18°C protected from light. The shelf life is 12 months tentatively. The number of repeated freezing and thawing should not be more than 4 cycles. It can be stably stored for 5 days below -18°C and protected from light during transportation.

[Requirements for Samples]
1.1 Stool Sample
Use a stool collector to collect stool samples from patients with suspected adenovirus infection in a disposable sterile container and close the tube cap tightly. Sample amount requirements:
Solid sample: The collection volume is about the size of a small ball with a diameter of about 2cm. Liquid samples: The collection volume is about 5mL.
Test samples should be stored at 2-8°C for no more than 7 days, and should be stored below -18°C for no more than 4 months, and could be stored below -70°C for long-term. The samples should be repeated freezing and thawing.
